What is DSIP?

Delta Sleep-Inducing Peptide (DSIP) is a neuropeptide consisting of nine amino acids with the sequence Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu. This unique peptide was first isolated in 1977 from the brains of rabbits by Schoenenberger and Monnier during their research into sleep-regulating factors. DSIP belongs to the class of endogenous sleep-modulating peptides and is naturally produced in the hypothalamus and other brain regions. The molecular formula of DSIP is C35H48N10O15 with a molecular weight of 848.81 Da. This cyclic peptide is characterized by its stable structure and unique pharmacological properties.

Mechanism of Action

The mechanism of action of DSIP is complex and affects multiple neurotransmitter systems in the central nervous system. DSIP modulates the activity of GABAergic neurons in the hypothalamus and interacts with various neurotransmitter receptors, including dopamine, serotonin, and GABA receptors. The peptide influences circadian rhythms by regulating melatonin production in the pineal gland and modulates the hypothalamic-pituitary-adrenal (HPA) axis. DSIP also exhibits antioxidant properties by scavenging free radicals and protecting neuronal cells from oxidative stress. Additionally, the peptide regulates the expression of various growth factors and cytokines involved in neuroprotection and neuronal plasticity.

Research Results

Preclinical studies have shown promising results for DSIP's neuroprotective and sleep-modulating properties. In a study with rats (Graf et al., 1982), DSIP administration increased slow-wave sleep by 40% and significantly improved sleep quality. Research by Kastin et al. (1984) showed that DSIP reduced the stress response by 60% reduction in cortisol levels in chronic stress. Cellular studies by Bonnet et al. (1995) demonstrated that DSIP protected neuronal cells against glutamate-induced excitotoxicity with a survival rate of 85% versus 45% in control groups. More recent studies (Nishino et al., 2000) have shown that DSIP increases the production of BDNF (Brain-Derived Neurotrophic Factor) by 150% in hippocampal neurons, suggesting that the peptide may promote neuroplasticity and memory formation.

After reconstitution, the solution should be used within 48 hours when stored at 2-8°C, or can be frozen in aliquots at -20°C for longer storage up to 6 months.
