What is NAD+?

Nicotinamide adenine dinucleotide (NAD+) is an essential cofactor found in all living cells and belongs to the class of nucleotide cofactors. This dinucleotide consists of two nucleoside molecules linked by their phosphate groups, comprising nicotinamide riboside and adenosine. NAD+ was first discovered in 1906 by British biochemists Arthur Harden and William John Young during their research into alcoholic fermentation in yeast cells. The molecule plays a crucial role as an electron acceptor in redox reactions and is fundamental for cellular energy production.

Mechanism of Action

NAD+ functions as a critical cofactor for over 400 enzymatic reactions in the human body. The molecule primarily acts via three main pathways: as a cofactor for dehydrogenases in glycolysis and the citric acid cycle, as a substrate for sirtuins (SIRT1-7) involved in gene expression and DNA repair, and as a substrate for poly(ADP-ribose) polymerases (PARPs) that detect and repair DNA damage. NAD+ accepts electrons and is reduced to NADH, generating energy for ATP synthesis. Additionally, NAD+ activates sirtuin proteins by cleaving off NAM (nicotinamide), resulting in deacetylation of histones and transcription factors, leading to improved mitochondrial biogenesis and cellular stress resistance.

Research Results

Preclinical studies have shown promising results for NAD+ supplementation. A landmark study published in Cell (2016) by Yoshino et al. demonstrated that NAD+ precursors increased the lifespan of mice by 10-15% and improved mitochondrial function. In C. elegans models, NAD+ supplementation resulted in a 25% extension of lifespan (Mouchiroud et al., Nature 2013). Cell culture studies show that increased NAD+ levels enhance SIRT1 activity by 40-60%, leading to improved DNA repair and reduced oxidative stress markers. Research on yeast cells showed a 30% improvement in mitochondrial respiration and ATP production when treated with NAD+ precursors (Belenky et al., Cell 2007).

Available Variants and Purity

Storage and Handling

Reconstituted solutions should be used within 48 hours when stored at 4°C, or can be frozen at -80°C for long-term storage up to 6 months. Avoid repeated freeze-thaw cycles to prevent degradation.
