What is Semaglutide?

Semaglutide is a synthetic peptide analog of naturally occurring glucagon-like peptide-1 (GLP-1), belonging to the class of incretin mimetics. This compound was developed by Novo Nordisk and is structurally based on human GLP-1, with specific amino acid modifications that enhance its pharmacological properties. The molecule consists of 31 amino acids and contains a fatty acid chain covalently bound to lysine at position 26, which ensures an extended half-life through binding to albumin. These structural adjustments make semaglutide particularly suitable for research into glucose regulation and metabolic processes.

Mechanism of Action

Semaglutide acts as a selective agonist of the GLP-1 receptor (GLP-1R), a G-protein coupled receptor primarily expressed in pancreatic islets, gastrointestinal tract, kidneys, and brain. Upon binding to the GLP-1R, semaglutide activates the adenylyl cyclase pathway, resulting in increased intracellular cAMP concentrations. This leads to glucose-dependent insulin secretion in pancreatic β-cells and inhibition of glucagon secretion in α-cells. Additionally, semaglutide delays gastric emptying by interacting with receptors in the hypothalamus and the autonomic nervous system. The peptide also modulates food intake by activating satiety signals via the arcuate nucleus in the hypothalamus, which has important implications for metabolic research.

In preclinical studies, semaglutide has demonstrated consistent effects on glucose homeostasis.
